Hi,
What type of account would I need to purchase to create sudomains under your 100webspace.net domain? ie.johnsdomain.100webspace.net
How many subdomains can be created per account?

The "Free" one is suitable for this purpose. You'll be able to create and use up to 5 subdomains on this hosting plan.

To clarify, I am looking to create volume subs on your 100webspace.net domain, and not on one of my own domains. Does the business plan truly offer unlimited subs on '100webspace.net'?

Further to my new question above, can I control the dns on your free subs,so as to point them at my server?

There should be no problem for you to redirect the subdomain names to point to your server. In case you have a dedicated IP address on your server you will be able to set it as an A Record for the subdomain names via your account control panel with us - Site Management > Custom DNS Records menu.
